How long does it take to connect Booking.com to a booking system?

On the Booking.com side you choose the connectivity provider and approve access. Then each of your room types and rate plans is mapped to the system's records so bookings land in the right place.
Airbnb connects in a similar way, by linking the listing. Existing future bookings are imported so the calendar is complete from day one.
